- Matthew Daddario was born and raised in New York to Richard and Christina Daddario, both lawyers. He studied business at Indiana University in Bloomington. He graduated in 2010, after which he began studying acting and started auditioning.
He is the middle child, with a younger sister, Catharine Daddario, and an older sister, Alexandra Daddario, who is also an actress.

- Originally auditioned for the part of "Jace Wayland" in the Shadowhunters (2016) TV series. When the book author, Cassandra Clare, watched his audition video, she tagged it "Would be perfect Alec" for the producers. Later, he auditioned and landed the role of "Alec Lightwood".
- His father, Richard C. Daddario, was a Prosecutor for the U.S. Attorney's Office in NYC for 13 years, before being named NYC Police Department Deputy Commissioner for Counterterrorism (2010-2013). His mother is lawyer Christina Titus.
